The nonrandom association between alleles at different loci is termed linkage disequilibrium. This note discusses the origin of this concept, its theoretical genetic basis, its estimation and tests of significance, and applications. Linkage disequilibrium studies are critical in human disease gene mapping and is now a major area of research with the availability of variation of the human genome sequence.
