The first section of this unit discusses basic principles of sterile technique, while the second section discusses medium preparation. The Basic Protocol describes subculturing of a monolayer culture grown in petri plates or flasks. Support protocols describe freezing of monolayer cells, thawing and recovery of cells, counting cells using a hemacytometer, and preparing cells for transport. An Alternate Protocol describes freezing suspension cells.
