Structure of RNA, its physical/chemicals properties and basic considerations in RNA analysis begin the discussion in this chapter. Methods of RNA extraction, particularly phenolic extraction and guanidium salt methods are described along with methods of purification, such as the removal of DNA from RNA. Examples of RNA isolation from nuclear and cytoplasmic cellular fraction are used as examples. RNA fractionation by chromatography and the RNA isolation from small number of cells are presented. Finally the assessment of quality, and quantitation of RNA are described.
