Abstract

We give an overview of the physical basics, experimental techniques, and selected applications of single molecule fluorescence in biophysical research. The photophysical principles underlying single molecule fluorescence resonance energy transfer (smFRET) experiments are treated. Its application in protein biophysics, in particular protein folding and conformational dynamics associated with enzymatic activity, is outlined with selected model systems.
