Summary

This chapter provides a general overview of the application of organocatalytic cascade reactions in natural product synthesis and drug discovery and highlights recent remarkable advances in this important field. It reviews a few organocascade reactions namely amine-catalyzed cascade reactions, brØnsted acid–catalyzed cascade reactions, and bifunctional base/brønsted acid–catalyzed cascade reactions. It shows how these reactions can significantly increase synthetic efficiency, bringing the synthetic chemistry much closer to the goal of ideal synthesis. Meanwhile, it is believed that the new concepts and strategies developed for cascade catalysis inspire chemists to design and develop more cascade reactions that can be employed in the rapid and efficient syntheses of complex natural products.
