Summary

Ectopic mineralization is a common response to tissue injury and systemic mineral imbalance, and can lead to severe clinical consequences when present in joints, tendons, heart valves, and blood vessels. It has been hypothesized that the mineralization of matrices in any tissue is normally controlled by the ratio of procalcific and anti-calcific regulatory molecules, such that abnormal deposition of mineral is avoided. Alterations in this ratio induced by injury, disease, or genetic deficiency are postulated to induce ectopic mineral deposition. Over the past few years we have developed in-vitro and in-vivo models of ectopic calcification in vascular tissues to investigate potential inducers and inhibitors of this process. In this chapter we will highlight recent findings in this area, focusing on the roles of phosphate and osteopontin in vascular calcification.
