Abstract

This chapter reviews the use of water as a green solvent in the manufacture of synthetic drug molecules. The review is separated into four categories:

  1. enzymatic processes;

  2. processes in which the product is isolated by pH adjustment to the isoelectric point;

  3. carbon–carbon bond-forming cross-coupling reactions in water; and

  4. pharmaceutical processes using mixed aqueous solvents.

The manufacturing processes of several major pharmaceuticals are discussed, including Crestor, Lipitor, Lyrica, Tamiflu, and Viagra.
