Figure 1. Effect of test drugs on survival of Sandhoff disease mice. (A) Survival curves and (B) percentage of increase in life expectancy are presented. Survival was significantly different in all treatment groups (p < 0.0001, n = 6 to 10) compared with untreated controls (n = 11). Survival in combination therapy groups was significantly different (vitamin E plus aspirin, p < 0.02; NB-DNJ plus indomethacin, p < 0.0001; NB-DNJ plus aspirin, p < 0.0001) compared with the individual monotherapy groups. The NB-DNJ plus indomethacin combination resulted in 6% synergy (p < 0.05) with a maximum improvement of a 66% increase in survival. The NB-DNJ plus aspirin combination therapy resulted in 11% synergy (p < 0.05) with a maximum improvement of a 73% increase in survival.
