Type II NKT hybridoma 14S.15.5D was activated by lyso-GalCer. CD1d-transfected APC were pulsed with lyso-GalCer or vehicle (DMSO) as described before, washed and co-cultured with 14S.15.5D hybridoma cells over night (Blomqvist et al. 2009, Eur J Immunol 39:1726). IL-2 in culture supernatants was measured by ELISA. Data indicate mean and SD of triplicate cultures and are representative of three experiments. Supplemental Figure 3. CD1d expression of APC. Cell lines were stained with anti-CD1d or control antibody. Upper plot: filled histograms (overlapping to the right) indicate CD1d expression, and empty histograms (overlapping to the left) show control antibody staining.
