SDS-PAGE and immunoblotting were performed on crude membrane fractions isolated from liver, kidney, duodenum, jejunum, ileum, and colon of mice. mOstB_91 antibody was used at a 1:1000 dilution alone (A) and in the presence of 50 mg of the peptide used to synthesize the antibody (B). Blots were then probed with a secondary antibody, antirabbit IgG horseradish peroxidase-linked F(ab′)2 fragment from donkey. For the antibody specificity analysis, 80 mg of protein was used per membrane fraction type, except for the ileum, in which 20 mg was used. Lane 8, as a positive control, is 0.5 mg of yeast secretory vesicles containing mOstb isolated fromS. cerevisiaetransformed with an expression vector containing the mouse Ostb gene.
