HEP_25667_sm_SuppFig1.tif3936KSupporting Information Figure 1. Woodchuck TGF-β1 (wTGF-β1) was cloned from the WHC17 hepatoma cell line. Woodchuck amino acid (aa) sequence was aligned with the protein sequence of the human, rat and mouse TGF-β1 molecules. The homology between the four species is significantly high. The TGF-β1 mature protein includes aa 290 to 390; we found that hTGF-β1 differs from wTGF-β1 in only one aa.
HEP_25667_sm_SuppFig2.tif342KSupporting Information Figure 2. The diminished IL-12 responsiveness observed in WHV chronically infected woodchucks with high viremia does no change over time. IFN-? expression by PBMCs after IL-12 stimulation without any additional in vitro stimulus such as WHV peptides covering CD8 T cell epitopes was analyzed by quantitative RT-PCR.
HEP_25667_sm_SuppFig3.tif2288KSupporting Information Figure 3. Combination treatment of WHV-chronically infected woodchucks with IL-12 and P17 peptide or CTX, or treatment with IL-12 alone do not modify significantly the expression of immuno-stimulatory molecules in the liver. Wβ2-microglobulin, wTAP, wCD3, wHeavy chain, wIFN-α and wCD8 hepatic expression levels were analyzed by quantitative RT-PCR in liver biopsies obtained prior to adenovirus injection and 60 days after the initial induction of IL-12.
