(A) RK3E cells stably transfected with the pBI-Rsf-1-V5/cyclin E1 inducible expression vector show protein expression as early as 6 h after gene turned on by replacing doxycycline-containing culture medium with regular culture medium. Western blot analysis shows induction of both Rsf-1 and cyclin E1 in the RK3E cells. (B) Co-immunoprecipitation using anti-cyclin E antibody revealed RSF/cyclin E1 complex formation 24 h after both genes turned on.
path4147-sup-0003-FigureS2.docWord document140KFigure S2. Co-up-regulation of Rsf-1, SNF2H, and cyclin E expression in ovarian carcinoma cells [21]. SKOV3 ovarian cancer line which expresses an undetectable level of endogenous Rsf-1 was engineered to express Rsf-1 controlled by a Tet-off system. Rsf-1 induction increases both SNF2H and cyclin E1 protein levels in a time-dependent fashion based on western blotting analysis.
