sc-11-0045_sm_SupplFigure1.pdf727KSupplementary Figure 1. Positive and negative controls for immuno-histochemistry in Figure 1. (1B: keratin 14, 1C: involucrin, and 1D: filaggrin.) Controls were performed on human neonatal epidermis. Negative controls were performed with omission of the primary antibody. Scale bar = 20 μm.
sc-11-0045_sm_SupplFigure2.tif91KSupplementary Figure 2. 82 ± 2% of CD44+ALDH+ human adult keratinocytes are keratin 14+. CD44+ALDH+ keratinocytes were isolated from adult human epidermis (n=2) by FACS. Cells were collected on glass slides and stained with FITC-conjugated keratin 14 antibody. Scale bar = 10μm
sc-11-0045_sm_SupplFigure3.tif2430KSupplementary Figure 3. Growth of CD44+ALDH+ and integrin α6hiCD71lo populations of cells in keratinocyte growth medium and in embryonic stem cell medium. These are the additional experiments to those shown in Fig. 7. Keratinocyte growth medium was Medium 154CF with human keratinocyte growth supplement (Invitrogen). Embryonic stem cell medium was ESC-SureTM DMEM medium (Applied StemCell). The integrin α6hiCD71hi population was included because it is considered a putative TAC population (Li, A., et al 1998 and Li, A., et al 2004).
