Fig. S1. A. Expression of Rab14 isoforms in Drosophila adults. Graph showing relative expression of the Rab14 isoforms RA, RB and RC in w1118 as measured by qRT-PCR.
B and C. Rab14 mutants show no defect in phagocytic uptake of E. coli. Wild type and Rab14 mutant adults were injected with fluorescein-conjugated E. coli followed by Trypan blue. Representative images and quantification of fluorescence intensity of wild type and the Rab14 mutant to examine the uptake of fluorescein-conjugated E. coli 30 min post injection. Each dot represents the intensity associated with one fly. The experiment was repeated 3 times. The data were analysed using an unpaired t-test.
D. Rab14 isoforms RA and RB shows similar localization in haemocytes. Haemocytes expressing Rab14RB-mRFP (magenta) and Rab14RA-YFP (green) driven by cgGAL4 were bled out from larvae and fixed. Images were obtained using confocal microscopy. The arrow indicates intracellular compartments showing colocalization. Scale bar, 5 μm.

Graph comparing relative expression of Rab14-RB in w1118 to Rab14 RB expression after hmlGAL4 or cgGal4 driven expression of UAS-Rab14RB transgene as measured by qRT-PCR.|

Haemocytes expressing Rab14RB-mRFP (magenta) and FYVE-GFP (green) driven by cgGAL4 were bled out from larvae and fixed. Images were obtained using confocal microscopy. Arrow indicates intracellular compartments showing colocalization. Scale bar, 5 μm.|
