B and C. Rab14 mutants show no defect in phagocytic uptake of E. coli. Wild type and Rab14 mutant adults were injected with fluorescein-conjugated E. coli followed by Trypan blue. Representative images and quantification of fluorescence intensity of wild type and the Rab14 mutant to examine the uptake of fluorescein-conjugated E. coli 30 min post injection. Each dot represents the intensity associated with one fly. The experiment was repeated 3 times. The data were analysed using an unpaired t-test.
D. Rab14 isoforms RA and RB shows similar localization in haemocytes. Haemocytes expressing Rab14RB-mRFP (magenta) and Rab14RA-YFP (green) driven by cgGAL4 were bled out from larvae and fixed. Images were obtained using confocal microscopy. The arrow indicates intracellular compartments showing colocalization. Scale bar, 5 μm.
