The complement system fulfils important functions in the host’s defence against bacterial infections, including direct bacterial killing via the Membrane Attack Complex (MAC or C5b-9). Gram-positive bacteria are protected from MAC-dependent lysis; yet several Gram-positive pathogens secrete proteins that inhibit C5b-9 formation. Here, we show that complement activation on Gram-positive bacteria results in specific surface deposition of C5b-9 complexes, occurring at specialized regions on the bacterial cell. This demonstrates a previously unrecognized interaction between the C5b-9 complex and Gram-positive bacteria.
