The plates are prepared by using 10 ml of 1% agar with 5 mg per ml freeze-dried M. lysodeikticus. 2 mm diameter holes are punched in each plate and 1 µl of exudate placed in each well. Following incubation at 33 °C for 24 h the diameter of the zone of inhibition is measured and lytic activity determined by comparison with a serial dilution of hen egg white lysozyme.
