      The xylene ring of riboflavin (vitamin B2) is assembled from two molecules of 3,4-dihydroxy-2-butanone 4-phosphate by a mechanistically complex process that is jointly catalyzed by lumazine synthase and riboflavin synthase. In Bacillaceae, these enzymes form a structurally unique complex comprising an icosahedral shell of 60 lumazine synthase subunits and a core of three riboflavin synthase subunits.

      Membrane-bound type prenyltransferases for aromatic substrates play crucial roles in the biosynthesis of various natural compounds. Lithospermum erythrorhizon p-hydroxybenzoate: geranyltransferase (LePGT1) responsible for shikonin biosynthesis is a model of such enzymes. We utilized a baculovirus-Sf9 expression system to generate a high-yield LePGT1 polypeptide, and efficient solubilization and purification procedures were developed, with which enzymatic characterizations were carried out.

      CAPE regulation of the expression of genes involved in cell cycle arrest and apoptosis was found to be associated with the increased expression of E2F-1. E2F-1 silencing abolished CAPE-mediated effects on cell cycle arrest, apoptosis, and related gene expression. Taken together, these results indicate a crucial role for E2F-1 in CAPE-mediated cellular activities in cervical cancer cells.

      Calpain-7, a mammalian ortholog of fungal PalB, is activated in the ESCRT system. We designed a non-physiological substrate by fusing calpastatin domain 1 with the Bro1 domain of ALIX and determined a cleavage site. Mutational analyses revealed importance of the N-terminal MIT domains and the C-terminal C2-domain-like (C2L) domains of calpain-7 to exert the limited proteolytic activities in HEK293T cells.

      Mutagenesis and kinetic studies identify a key active site residue in the α-d-phosphohexomutase superfamily. Histidine is strongly conserved at this position in many proteins, while lysine is also often conserved at the structurally corresponding position in other family members. Structural analyses suggest that this residue may serve as the general base in the phosphoryl transfer reaction.

      We investigated the role of Cap1p in apoptosis of Candida albicans and demonstrated that more apoptotic cells, more necrotic cells, decreased mitochondrial membrane potential and intracellular ATP level accompanied with increased caspase activity and ROS accumulation occurred in the cap1Δ/Δ mutant upon apoptotic stimuli. The cell fate of cap1Δ/Δ was, as least partly, caused by decreased glutathione and GLR1 expression.

      We have examined the potential role of Gdf6 in adipogenesis of mesenchymal stem cells (MSCs). overexpression of Gdf6 committed pluripotent mesenchymal C3H10T1/2 cells to an adipocyte lineage. Type I receptor Bmpr1a and type II receptors Bmpr2 and Acvr2a mediated Gdf6/Smad and Gdf6/p38 MAPK signaling pathways. Downregulation of Runx1t1 was required for Gdf6-induced adipocytic commitment.

      In this paper we showed that some antibiotics and their complexes with cupric ions inhibiting the delta ribozyme could bind to different regions or bind in a different way. However, their high binding affinity to the ribozyme seemed to be of primary importance for effective inhibition.

      The proteasome inhibitor MG-132 belongs to the group of peptide aldehydes. Its toxicity was shown in multiple reports. Here we show that MG-132 up-regulates the expression of monocyte chemotactic protein-1 induced protein 1 (MCPIP1) by the activation of MCPIP1-coding gene. We present the requirement of ERK and p38 in this effect and postulate the involvement of MCPIP1 in MG-132 toxicity.

      Ensemble FRET experiments reveal significant electrostatic repulsion between the globular domains of calcium-free calmodulin, site-specifically labelled using expressed protein ligation. At low salt, the protein has a relatively extended conformation and the distance between the domains is further increased by denaturation, by heat, or by non-ionic denaturants. The repulsion between domains is screened by salt or diminished by calcium binding.

      N-terminal domain of Pyrococcus furiosus l-asparaginase forms stable, polydispersed oligomers. It helps in folding of the C-terminal domain and prevents thermal and refolding-mediated aggregation of a variety of substrate proteins, thus acting as molecular chaperone. It also inhibits polyQ and Aβ amyloidogenesis. It may be used in aggregation prevention in heterologous expression systems and also in therapeutic intervention of amyloidogenesis.

      After EGF stimulation, PPM1A negatively regulated ERK phosphorylation at early time points. Biochemical results proved the direct association between PPM1A and phospho-ERK. Further kinetic studies identified the interaction between the R186 of PPM1A and the E203 and pY204 of phospho-ERK as a hot-spot for PPM1A/phospho-ERK interaction.

      Mutations of several spliceosome-associated proteins from yeast do not affect pre-mRNA splicing. We show that splicing defects can be detected in many such mutants by using a set of pre-mRNA reporters with a wide range of splicing characteristics. For example, deletion of a proline-rich region at the N-terminus of Prp8 induces alternative usage of a duplicated branchpoint/3′ splice site region.
