Figure S1B. BIRB 796 has minimum effect on MM cell viability. (A) MM.1S, RPMI8226, OPM1, and U266 MM cell lines were cultured with control media or BIRB 796 (100-800 nM) for 72 h. Cell growth was assessed by MTT assays; data represent mean (± SD) of quadruplicate cultures.

Figure S1C. MM.1S cells were cultured with 400 nM BIRB 796 for 12 h; 25 µM Z-VAD (Z-VAD-FMK: Calbiochem, San Diego, CA, USA) was then added for an additional 1 h, followed by control media (clear bars) and 5 (grey bars) or 10 nM (black bars) bortezomib for the last 24 h. Cell growth was assessed by MTT assays, and data represent mean (± SD) of quadruplicate cultures.

Figure S1D. BIRB 796 does not change expression levels of Hsp90. MM.1S cells were cultured with control media or 400 nM BIRB796 for 24 h. Total cell lysates were subjected to immunoblotting using anti-Hsp27, phospho-Hsp27 (p-Hsp27), Hsp90, and alpha-tubulin antibodies.
