Fig. S1. Correlation between expression levels of Preso and the extent of dendritic outgrowth. Expression levels (fluorescence intensities in arbitrary units) of Preso in neurons transfected with sh-vec, sh-Preso, and sh-Preso + rescue were plotted against the total lengths of dendrites. The linear fit in dark brown is for neurons transfected with sh-vec or sh-Preso (R = 0.61, P < 0.0001). Higher Preso expression in neurons transfected with sh-Preso + rescue showed insignificant correlation with dendritic outgrowth (blue line; R = 0.16 for rescue, P = 0.31), suggesting that expression above the physiological levels does not stimulate additional dendritic outgrowth.

Fig. S2. The lack of effect of Preso overexpression is not due to an insufficient present of βPix. (A) Effects of the co-overexpression of Preso WT and βPix on dendritic outgrowth. Cultured neurons doubly transfected with EGFP-C1 or EGFP-βPix with Flag-Preso WT (DIV 9-16) are visualized by Flag and EGFP antibodies. Scale bar, 100 μm. (B) Quantification of total length of dendrites (n = 17 for WT+EGFP-C, n = 18 for WT+EGFP-βPix constructs, P = 0.8303, Student’s t-test). Histograms show mean + SEM.
