Figure S2: Cumulative mating success rate (%) of wild-type males without wings and pain1 mutant or wild-type females. We observed mating behavior for 20 min. The mating success rate defined as the percentage of pairs that copulated was determined every 1 min. The wings of CS males were removed with microscissors during 3 min cold anesthesia (< 4°C), and cold-anesthetized CS males with wings were used as controls after they recovered from anesthesia. A total of 68 pairs of wingless males and CS or pain1 females were observed, and 20 pairs of winged males and CS or pain1 females were observed

Adult female brains were fixed with phosphate-buffered saline containing 4% formaldehyde for 20 min at room temperature.
