Human and mouse embryonic stem cells (ESCs) are pluripotent and can give rise to any adult cell type. hESCs are classified as “primed” and one step differentiated from their “naïve” mESC counterpart. We review the strategies used to generate pluripotent cells and discuss the elusive derivation of naïve human ESCs.
