This work describes the establishment of a Sf9 cell line using flipase-mediated cassette exchange for site-specific gene integration. After stable integration of the tagging cassette (expressing dsRed), successful exchange was achieved for several clones displaying similar reporter EGFP expression to that of the tagged cell line. This predictability demonstrates the usefulness of this new Sf9 master cell line as a re-usable platform to produce multiple recombinant proteins, skipping extensive clone screening, and as an alternative to baculovirus infection.
