Room for errors: HIV-1 reverse transcriptase is remarkable for its low fidelity. This report describes a series of non-natural nucleotides used to explore the biophysical mechanism(s) accounting for error-prone replication catalyzed by this polymerase. This data supports a mechanism highlighting specific interactions of the incoming nucleotide with amino acids capable of mediating hydrogen-bonding interactions.
