Black is beautiful! The azo-coupling reaction between Methylene Violet 3RAX and functionalized tertiary anilines has been revisited to obtain a series of novel BHQ-3 dyes with various reactive handles (see figure). The bioconjugation ability of such modified dark quenchers was illustrated by the preparation of urokinase-sensitive fluorogenic probes in good yields.
