The high sensitivity of fluorescence correlation spectroscopy (FCS) enables measurements of hydrodynamic radius of single chain of polyelectrolyte at low salt condition, which has been a challenge for conventional dynamic light scattering (DLS). In this study, a direct comparison was made between these two methods on a modeled polyelectrolyte system, poly 2-vinylpyridine, and the results demonstrate the great advantage of FCS in the study of polyelectrolytes.
