Two tracers 89Zr-labeled cetuximab (89Zr-Df-Bz-NCS-cetuximab) and 89Zr-cetuximab conjugated to gold nanoparticles (AuNPs-PPAA-cetuximab-89Zr) were injected in A431 xenograft-bearing mice and their biodistribution was studied using quantitative PET imaging. Radioimmunoconjugates were carefully characterized before injection. Immuno-PET studies revealed no significant difference in tumor uptake for AuNPs-PPAA-cetuximab-89Zr up to 72 h after injection, compared with 89Zr-Df-Bz-NCS-cetuximab and showed a maintaining of high tumor contrast. However, the liver uptake of AuNPs-PPAA-cetuximab-89Zr was higher. In vivo blocking experiments demonstrated selective tumor targeting after coupling reaction.
