In this work the sensitivity of DA364 to detect αVβ3 integrin-overexpressing tumors was evaluated. DA364 derives from the conjugation of Cy5.5 with a RGD probe selected among a collection of cyclic peptidomimetics. DA364 displays high binding affinity for αVβ3 integrin and prolonged visualization of glioblastoma xenografts in mice. Its specificity was confirmed by inhibition of tumor uptake in the presence of nonfluorescent RGD peptides, suggesting that DA364 is a promising optical imaging tool for tumor diagnosis and guided surgery.
