Three-dimensional imaging with sub-diffraction resolution is achieved by improving the excitation conditions for saturated excitation (SAX) microscopy. Optimal modulation conditions that provide sufficient fluorescence saturation and little photobleaching are determined both theoretically and experimentally, thus enabling the volumetric sub-diffraction imaging of actin cytoskeleton in a cell.
