Sensitive detection of synthetic 1,2-dimyristoyl-3-trimethyl-ammonium-propane (DMTAP) lipid employing drop coating deposition Raman (DCDR) and surface-enhanced Raman scattering (SERS) from dried droplets is reported. Reproducible spectra of ~10 μM DMTAP initial concentration were measured by DCDR without considerable loss of its solution properties. SERS spectra of DMTAP were obtained down to ~0.3 μM which is about five orders of magnitude lower than that of conventional Raman spectroscopy. In this case, the solution properties of DMTAP were perturbed by adsorption on the Ag nanoparticles.
