To our knowledge, our manuscript is the first time to report the accurate role of autophagy in high-LET radiation-induced cytotoxicity to tumor cells. High-LET carbon ions elicited autophagy in tumor cells effectively via depressing the PI3K/Akt pathway. Manipulating autophagy could alter the cellular radiosensitivity of tumor cells and inhibiting autophagy enhanced the high-LET radiation induced apoptotic cell death, leading to an increase in radiosensitivity.
