The Na+/H+ exchanger (NHE1) requires cellular ATP for its activity, despite no requirement of energy input from ATP hydrolysis. Using photoaffinity labeling technique on the purified complex protein of NHE1 and its cytosolic subunit CHP1, we obtained evidence that NHE1 is an ATP binding protein. Thus, ATP may serve as a direct activator of NHE1.
