Type IA DNA topoisomerases from bacteria are potential drug targets for which inhibitors are being discovered now. We identified a monoclonal antibody, 2F3G4, which specifically inhibited mycobacterial topoisomerase I in nanomolar range. The antibody stimulated enzyme-mediated DNA cleavage and elevated protein-DNA covalent adducts. The mode of inhibition opens up avenues to develop new, small molecule inhibitors for bacterial topoisomerase I.
