The dual-specificity phosphatase SEX4 is required for reversible glucan phosphorylation during starch degradation in plants. We demonstrate that SEX4 activity is modulated between reduced (active) and oxidized (inactive) states by a disulfide linkage with the catalytic cysteine, like other redox-regulated phosphatases. This represents the first plant phosphatase to undergo reversible oxidation via disulfide bond formation like its mammalian counterparts.
