Intestinal organoids grow from single Lgr5-positive stem cells. We introduce a single cell-based computer model of these systems. In simulation studies, we find that proliferation-induced shape fluctuations are sufficient to induce crypt-like domains. Moreover, assuming that stem cell differentiation is triggered by local tissue curvature, stem cell expansion within the organoids sensitively depends on tissue biomechanics
