Spinosad is a macrocyclic lactone insecticide that acts on nicotinic acetylcholine receptors (nAChRs). We have cloned the nAChR α6 subunit from the western flower thrips Frankliniella occidentalis (Foα6) and have identified a single transmembrane point mutation that is associated with resistance to spinosad. Characterization of the mutation in recombinant nAChRs is consistent with spinosad having an allosteric mechanism of action.
