The neurotrophin BDNF promotes peripheral myelination via the neurotrophin receptor p75NTR. To selectively target p75NTR, we have generated a small peptide (cyclo-dPAKKR) that structurally mimics the region of BDNF that binds p75NTR. Here we have demonstrated that cyclo-dPAKKR promotes peripheral myelination in vitro and in vivo, suggesting that selective targeting p75NTR, via cyclo-dPAKKR, is a strategy worthy of further investigation for the treatment of peripheral demyelinating diseases.
