As regulators of vesicular transport, Rab GTPases are prime candidates to function in the trafficking of virus glycoproteins. We have found that Golgi-associated Rab6 is crucial for herpes simplex virus 1 glycoprotein transport from the Golgi to the plasma membrane, a process that involves the Rab6 effector ERC1, and the redistribution of Rab6 positive vesicles to the cell periphery. This Rab6-specific step is essential for virus production, indicating glycoprotein transport to the plasma membrane is a prerequisite for envelopment.
