The cover picture shows a model of an integrin-mediated signaling event. An intracellular protein binds to the integrin in the ground state (left). This leads to a conformational change, which exposes an extracellular binding site. Binding of a ligand on the extracellular site (middle) triggers a further conformational change, which in turn may be the starting point for an intracellular response (right). An X-ray structure of the activated form allowed modeling and docking studies of the ligand binding event. The computational studies demonstrate that the X-ray structure is only a snapshot in a multistage process. The new three-state model of integrin signaling is in agreement with biochemical data as well as other computational studies of the transmembrane domains.
