The change in fluorescence anisotropy depends on rotational diffusion of the fluorophore. Protein–fluorophore interaction causes the fluorophore to rotate more slowly and leads to an increase in fluorescence anisotropy, as shown by M. Y. Berezin et al. on p. 716.
