In this issue's Minireview, McGuigan et al. describe prodrug technologies aimed at delivering nucleoside monophosphates into cells, with applications in anticancer and antiviral therapeutics. A new target for the nonsteroidal anti-inflammatory drug sulindac is highlighted by Rauh and Simard, who describe the implications of repurposing sulindac.
This issue's thematic cluster is focused on computational medicinal chemistry and includes two Communications and five Full Papers. From the virtual screening efforts described by Spänkuch and colleagues, who use a homology-based structural model of human Plk1 to identify a selective Plk1 inhibitor, to the introduction of selectivity cliffs, which are identified through a computational analysis of structure–selectivity relationships in a systematic and quantitative manner, by Bajorath et al.
