Roots elongate by adding new cells at the tip, similar to constructing a chimney by piling bricks on top of one another. The newly-formed cells gradually change the size and outer shape of the root, as well as its inner organization. But what genes control this process of cell differentiation, and what triggers it in the first place? These are important questions to be answered, which is why I nominated this contribution by Huang and colleagues as a Featured Article in The Plant Journal.

The paper reports a key gene involved in the control of the specification of root outer cell layers, as well as the formation of root hairs and lateral roots, in rice. The gene, named Docs1, encodes an enzyme that is a member of the leucine-rich receptor-like kinase (LRR RLK) family. Plant and animal cells carry large numbers of these LRR RLKs, which are believed to play a role in receiving signals from the outside environment and the subsequent modulation of gene expressions. In this context, it is expected that certain stimuli received by this LRR RLK could trigger the process of cell differentiation.

The authors were able to demonstrate polarized localization of this LRR RLK on the cell membrane, which is an interesting and important finding because it provides an indication as to the possible orientation of the incoming signal(s), although the nature of the signal(s) and the downward signaling systems remain unknown. Also, by comparing the roots of wild-type and knock-out mutant plants, results are reported that suggest Docs1 might directly or indirectly regulate many other genes involved in the development of root outer cell layers in rice. Overall, the identification by these authors of a key gene involved in the root cell differentiation system is a major contribution to this emerging story.
