      Chlamydiae are obligate intracellular pathogens representing a serious public health concern worldwide, and yet, there is no vaccine available. Chlamydial cell surface molecules that are essential for the adhesion and internalization process are good targets for intervention strategies; however, there is little known about the molecules involved. Here, we describe the novel chlamydial surface protein CPn0473 that mediates adhesion to human cells via a specific stretch of 50 amino acids in the C-terminus. We show that CPn0473 is involved in an early step of internalization of C. pneumoniae which occurs via specific domains, named lipid rafts, mediated by the N-terminal part of the protein. Thus, CPn0473 is the first cell surface protein found in Gram-negative bacteria that interacts with lipid-rafts-associated molecules in the plasma membrane of the host cell to promote the uptake process. The identification of CPn0473 as a surface-localized chlamydial protein, which is essential for the internalization process and thus for the establishment of the infection, has also implications for the development of novel anti-chlamydial therapeutics. For example, targeting the ability of CPn0473 to bind to lipid raft microdomains of the host cell plasma membrane might prove a useful strategy to diminish the virulence capacity of C. pneumoniae.
