Lipids

Virtual issue cover legend: To promote the formation and accumulation of lipid droplets cells were incubated with a medium that contained fatty acids. The presence on droplets of caveolin-2 (red) and TIP-47 (green) was analysed with specific antibodies (top panel). A combined targeting motif which includes a hydrophobic domain followed by a basic sequence (HyD-End3) mediates sorting of otherwise cytosolic proteins such as green (bottom left) and orange (bottom right) fluorescent proteins to lipid droplets (see details in Ingelmo-Torres, et al. Traffic. 2009 Dec 10:1785-801).

Lysosomes

Virtual issue cover legend: Melanosomal resident protein Tyrp1 labeled in green, the late endosome/lysosomal resident protein LAMP1 labeled in red, and the early sortiong/ recycling endosomal tSNARE Syntaxin 13 labeled in blue. As shown in the image, none of the markers overlap - indicating that melanosomes are distinct from early and late endosomes/ lysosomes. Image courtesy of Mickey Marks and Subba rao Gangi Setty.

Virtual Issue Nuclear Transport

Nuclear Transport

Schematic model for nuclear protein import. Carrier binds to cargo in the cytoplasm, docks at the NPC and is translocated to the nucleus where RanGTP binds to the carrier dissociating the cargo-carrier complex. The carrier-RanGTP complex is then exported to the cytoplasm where RanGAP and RanBP1 activate the RanGTPase activity and dissociate the carrier-Ran complex, thus freeing the carrier for a further cycle of nuclear import.
